Cell based assays, while originally used mostly for secondary screening, are now increasingly being used for primary screens. This has been tremendously valuable in screening all types of compounds and particularly those compounds for druggable cell membrane proteins such as GPCRs, ion channels and cell receptor kinase proteins. It is well understood that the success of the screening campaign is only as good as the reagents used in the assays.
The Selexis SURE Cell line development™ technology can be used to generate cell lines for a whole range of cell based assays, including enzyme and cell receptor activity assays. Cell lines created with the Selexis Genetic Elements™ (SGEs) express recombinant proteins at significantly higher levels than those obtained with cell lines generated using traditional methods and exhibit higher stability. As a result, it is now possible to generate stable cell lines expressing difficult-to-express biomarkers such as GPCRs, ion channels and cell receptor kinases at levels that are assayable.
Cell-based assays built around the Selexis SURE Cell line development™ technology have the advantages of:
- High expression increasing the chances of generating cell lines with robust signals
- Expression stable over >60 generations making it unnecessary to keep thawing fresh batches of cells
- Reduced chances of failed screening runs due to loss of target expression in the assay
- Selective media is not required to maintain the high-level expression
