In most cells > 80% of genes are switched off or silenced due to tightly packed chromatin structure. A major challenge of recombinant protein expression is the successful integration of recombinant genes into permissive areas in the chromatin where transcription is NOT silenced as well as avoiding epigenetic gene silencing. Selexis Genetic Elements™ are novel DNA-based elements that control the dynamic organization of chromatin within all mammalian cells and allow for higher and more stable expression of recombinant proteins. The SGEs function by insulating nearby genes from the silencing effects of surrounding chromatin thereby increasing copy number-dependent and position-independent expression of genes. Cell lines created with SGEs express recombinant proteins at significantly higher levels than those obtained with cell lines generated using traditional methods and exhibit higher stability when cell lines are cultivated in absence of any drugs for long-term periods.
